Custom Directories aren't working

For some reason I can not get custom directories to work for me. I've gotten them to work on my other computer, so I know I'm setting them up correctly. I've tried fresh installs and it doesn't do a thing. I have no idea what unique information I can provide to help identify the problem, so if someone could tell me what to provide, that'd be great.
